━━━━━━━━━━━

How can we accurately and reproducibly identify different tuna species? Students receive tuna specimens (fillets or sushi) and analyze them using molecular biology techniques to determine species identity. The course culminates in mini-presentations of results, designed to: (1) master fundamental experimental skills and scientific discussion methods, and (2) understand life science concepts like DNA and PCR commonly referenced in daily news. Experimental Workflow: ① Course overview and methodology explanation ② DNA extraction from tuna specimens ③ DNA amplification via PCR ④ Electrophoresis and fragment detection ⑤ Sanger sequencing of PCR products ⑥ Species identification through database comparison ⑦ Lab meeting-style presentation and discussion Students perform DNA extraction, PCR amplification, Sanger sequencing, and sequence analysis. Final DNA sequences are compared against known tuna databases to identify the specific tuna genus species provided. ━━━━━━━━━━━ Part B | Independent "Thinking Training" Projects ┄┄┄┄┄┄┄┄┄┄┄ Beyond the structured workflow, students receive individualized assignments tailored to their learning progress and experimental advancement. These challenges cannot be easily resolved through web searches, generative AI, or reference materials—requiring students to design and conduct original experiments (e.g., testing DNA extraction methods and performing PCR from other biological or food samples). This "thinking training" component adds open-ended challenges to the prescribed practical flow, providing experience with problems lacking predetermined solutions. We hope that you will enjoy your stay on this course.

A. Lectures [All Students] Instructor-led explanations of key concepts and methodologies. B. Laboratory Notebook Maintenance [Individual] Students maintain detailed experimental records in designated paper notebooks or electronic PDF formats. Instructors review and provide feedback on notebooks after each session. C. Experimental Work [Individual] Hands-on sample handling and data collection. D. ADVES Problem Book Assignments [Individual or Group] Progress-based assignments designed to deepen practical understanding and provide "thinking training." Students may work individually or collaboratively using laboratory computers, personal devices, or Komaba campus library resources. E. Data Analysis and Discussion [Individual or Group] Students analyze experimental results independently or collaboratively, utilizing available computational resources on campus or personal devices. F. Lab Meeting-Style Presentations [Individual or Group] Students present experimental results and analyses without formal slide preparation. Raw laboratory notes are projected directly using overhead projectors, with hybrid participation (in-person + online) available. Two Presentation Approaches: (1) Raw Data Sharing Students photograph and share unedited laboratory notes via camera or smartphone. This approach emphasizes authentic data sharing, increases error detection through peer review, and provides valuable collaborative feedback. (2) Formal Slide Presentations Students create polished presentations (PowerPoint, Google Slides, Keynote, PDF) highlighting key findings while omitting extraneous data. This format may be more accessible to unfamiliar audiences but risks masking analytical errors due to selective data presentation. 💡 Both approaches develop essential academic skills: authentic scientific communication and formal presentation techniques—balance between these methods enhances overall academic performance.
